The reason is due to a high-energy laser light can release energy at a short time and lead to selective destruction of the absorbing tissue. In the clinical and cosmetic medicine, laser light destroies melanin or other chromophores without causing change in the adjacent tissue by the concept of selective photothermolysis. However we can often find case reports with side-effects after laser therapy, such as scarring, purpura and post-inflammatory hyperpigmentation, which relate to the non-optimal parameters of laser setting. In order to improve the side-effects, the study attempt to use ultra-short laser pluse with wavelength 800nm and pulse duration between 40 fs (1 fs=10-15 sec) to 160 ps (1 ps=10-12 sec), in vitro test. We try to find out the lowest fluence and appropriate pulse duration that can carry out photothermolysis efficiently, and then assess the probability of applying this technique for photobleaching of pigmented skin.
In chapter 1, we review the history and fundamental theories about medical laser technique, and describe the motive of this study. In chapter 2, we introduce the biological, physical and chemical properties of melanin; in addition, we examine properties related to the laser-tissue interactions. In chapter 3, the preparation of our samples, experimental setup and the methods of data analysis are described in details. The results and analysis of our experiment are shown in chapter 4. In chapter 5, we discuss the results. The last chapter is the conclusion of this thesis and the prospective of future experiment.
