Protective Effect of Astaxanthin on Hydrogen Peroxide-Induced Cytoxicity in PC12 Cells. / 蝦紅素對過氧化氫誘導PC12細胞毒性之保護效果

The carotenoid pigment astaxanthin has important applications in the nutraceutical, cosmetics, food and feed industries. Astaxanthin is a strong coloring agent and a potent antioxidant .In this study we examined whether astaxanthin had a neuroprotection against H2O2-induced injury in PC12 cells. Calcium and lipid peroxidation play important roles in oxidative stress-induced cellular injury and apoptosis,which ultimately cause cell death. Pretreatment of PC12 cells with astaxanthin improved the cell viability, decreased malondialdehyde level and attenuated the intracellular Ca2+ concentration, and the reduced NO production in the H2O2-induced injury in PC12 cells.Furthermore, the [Caspase-3/pro-Caspase-3] and [Bax/Bcl-2] ratios were slightly low in H2O2-induced injury in PC12 cells after astaxanthin treatment . In summary, this study demonstrates that astaxanthin is able to relieve H2O2-induced oxidative stress and apoptosis in PC12 cells, at least in part, by Ca2+ antagonism and antioxidant mechanisms.
