Effects of Maternal Cadmium on Development and Metallothionein Expression in Zebrafish (Danio rerio) / 母方鎘對斑馬魚發育與金屬硫蛋白表現之研究

碩士 / 國立嘉義大學 / 水生生物科學系研究所 / 97 / Cadmium, a heavy metal, is known to influence the offspring through maternal transfer. Most studies focus on the maternal cadmium influences the relative heavy metal tolerance and production of offspring. Researches on the physiological mechanism of the infecting-cadmium parental generation to offspring and influence on the specific gene expression were nevertheless numerous. Therefore, the aims of this study were to confirm the effects of different doses of cadmium treatment on female and its offspring, and development of zebrafish (Danio rerio) larvae. The results of the experiment showed that cadmium decreased the parental mating success rate by 30 %. However, there is no significant difference among groups with regard to the number of spawned offspring. Cadmium contents, metallothionein and MT mRNA expression showed a positive correlation upon cadmium exposure. In addition, in female fish exposed to cadmium for 72 hours and immediately transferred to clean water, Cd contents of the ovary were not evidently reduced. The effect of cadmium on larvae development can be observed during 6-somite stage wherein growth delay occurred. In addition, the distance of ceratohyal head bone of larvae is also showed a dose response upon the maternal-Cd treatment. The hatching rate and survival rate of larvae from high maternal-Cd were not statistically significant. However, it seems to lead to early larvae hatching time and reduced hatching rate. Larvae exposed to 53.4μM Cd for 48 hours had evidently enhanced Cd tolerance. On the contrary, pericardial edema occurred in several larvae until 96hpf from high maternal-Cd. The maternal-Cd contents and MT mRNA expression were concentration-dependent. Based on these results, we suggest that cadmium, via maternal effect, influences the reproduction ability of female fish and causes abnormal development of offspring.
