碩士 / 國立高雄海洋科技大學 / 水產食品科學研究所 / 97 / Freshwater clams (FC), Corbicula fluminea, are a popular seafood and traditionally used as a Chinese remedy for liver disease and chronic hepatitis. The goal of this study was to identify the active compounds in freshwater clam, which are responsible for anti-hepatic fibrosis. The meat of FC was extracted with ethanol (FC-EtOH) and the ethanol fraction was further extracted with ethyl acetate (FC-EtOH-EA).
In vitro study showed that FC-EtOH and FC-EtOH-EA inhibited TGF-beta1-induce alpha-smooth muscle actin (alpha-SMA) secretion and collagen deposition in HSC-T6 cells by blocking Smad2/3 phosphorylation and nuclear translocation. The anti-hepatic fibrosis effect of FC-EtOH was also performed in vitro. From Masson’s trichrome stain, FC-EtOH significantly reduced the fibrosis scores of livers induced by DMN administration in Sprague-Dawley rats (SD rats). This study suggested that both FC-EtOH and FC-EtOH-EA exhibit anti-hepatic fibrosis effects in vivo and in vitro. Moreover, FC-EtOH-EA was subjected to separation and purification using silica gel column chromatography.
The compound,TNHD, showing strong anti-hepatic fibrosis was identified by NMR and LC/MS.
The anti-fibrotic effects and mechanisms of action of TNHD might provide suggestions in drug development and clinical application.
