碩士 / 國立臺灣海洋大學 / 水產養殖學系 / 97 / Small abalone(Haliotis diversicolor supertexta), is an important aquaculture species in Taiwan. Recently, the species has been exposted to China and South-Eastern Asian countries as a major cultured abalone. However, outbreaks of mass mortality in spat, juvenile and adult abalone due to unknown causes occurred in Taiwan science 2001.The shortage in spat supply and foot muscle atrophy syndrome in cultured abalone together with mass mortality threaten the industry and have not been well resolved until now.
Vibrio alginolyticus was found to be the dominant species in the environment of spat settling and was virulent to juvenile abalone. The addition of yolk IgY against V.alginolyticus to the spat settling system could enhance survived rate in a preliminary study.
In the present study, sterile sea water was found useful in the preparation and storage of injected abalone tissue for further virulent tests comparing to the ordinary buffer.
Virus-like particle(12.5-23 nm) could be observed in the specieman sample from diseased abalone during outbreak of mass mortality in Tou-Chen, I-Lan, by using negative stain in TEM. The filteried homogenates(0.22 μm) of tissue from diseased abalone could cause mass mortality after challenge.
Virus-like particles were obtained after sucrose-zone centrifuge of the same filtered homogenates as mentioned above. In addition these virus-like particles were virulent to the abalone causing 100 % mortality.
