碩士 / 國立臺灣海洋大學 / 食品科學系 / 97 / In this study, we show the antioxidant activity of ten peptides isolated from mackerel hydrolysates and found the tetrapeptide Val-Trp-Trp-Trp which shows the significant antioxidant activity than other peptides in mackerel hydrolysates can prevent LDL oxidation induced by copper ion. Furthermore, we demonstrated Val-Trp-Trp-Trp anti-atherosclerosis bioactivity using cell culture model. Val-Trp-Trp-Trp inhibit formation of foam cell through decreasing gene expression of scavenger receptor A and iNOS induced by ox-LDL in RAW 264.7 macrophages. However, reactive oxygen species (ROS) and activation of nuclear kappa B factor (NF-����) are the major risk factors in vascular disease. Val-Trp-Trp-Trp can reduces oxidative stress and prevents NF-���� activation as well as down-regulate inflammatory gene expression such as COX-2, CD36, IL-1, IL-8 induced by advanced glycation end products in RAW 264.7 macrophage. Val-Trp-Trp-Trp antioxidant activity may due to its multiple conjugated bonds and amino acid sequence. We demonstrated antioxidant and anti-inflammation function of Val-Trp-Trp-Trp through reducing cellular oxidative stress. A detailed understanding of this anti-inflammatory mechanism should be further investigated.
