碩士 / 國立臺灣科技大學 / 化學工程系 / 97 / The purpose of this research is (1) to investigate the effect of environmental factors such as light intensity, and chemical factors such as glucose and NaHCO3 on the growth of cyanobacteria of Synechocystis sp. PCC 6803 and (2) to study the expression of Enhanced Green Fluorescent Protein and Vitreoscilla Hemoglobin. Synechocystis sp. PCC 6803 grew well under photoautotrophic condition at light intensity of 34 μE m-2 s-1。The glucose can be used as carbon source with optimal concentration of 5 mM for culturing Synechocystis sp. PCC 6803. The cell growth was enhanced by employing glucose as carbon source. Its doubling time was about 40 % shorten than that cultured without glucose. The optimal NaHCO3 concentration was found to be 20 mM. Plasmids containing EGFP and VHb gene was constructed and transformed into Synechocystis sp. PCC 6803. The expression level of these two proteins was too low to be measured by SDS-PAGE. However, they can be detected by western blot. The expression of VHb had no obvious effect on cell growth and the rate of oxygen evolution. In contrast, adenosine triphosphate (ATP) content increased about 58% in stationary phase as compared with the wild type strain.
