Microalgae have drawn more and more attention as a promising source of biodiesel, so that rapid screening for algae strains with enhanced lipid production becomes essential.
The main motivation of this study is to evaluate the feasibility of using flow cytometry for rapidly detecting the oil content in cultivated Chlamydomonas reinhardtii. Traditionally, the lipid content in microalgae samples was determined using extraction and weighing procedures, which would take at least hours. The lipid of a microalgal cell can be stained with Nile red, a fluorescent dye that emit orange-yellow light when excited with a proper light source. With a properly designed set-up, flow cytometry is capable of measuring the lipid content of every individual cells in a microalgal population in liquid suspended form. Comparisons between flow cytometry and traditional extraction procedures in determining lipid content were carried out in this study.
This research method into three majority:The first part is the micro algae culture,Select test strains of micro algae as Chlamydomonas CC400,Microalgae Culture work including plate culture、sub culture、Scale up to the desired volume of culture.The second part is Including optical density (OD value) analysis, cell count, fluorescence microscopy and flow cytometry fluorescence emission intensity analysis, etc.The third part is sampling, centrifugal, freeze-dried, extraction of oil.
A high correlation (R2 > 0.97) between the Nile Red fluorescence intensity measured by flow cytometry and total lipid content assayed by the traditional lipid extraction method was observed for this microalgae. Our data have implied that this method is a useful tool and a quick technique for the screening of microalgal strains for lipid production.
