Functional Study of zebrafish zfp598 by morpholino anitisense oligonucleotides / 運用反義核酸進行斑馬魚zfp598功能之研究

碩士 / 國立嘉義大學 / 獸醫學系研究所(Graduate Institute of Veterinary Medic / 98 / Abstract
ZFP598 is belonging to C2H2-type zinc finger that also called Ring
finger protein. The ring finger motif is a protein-protein interaction
domain which has been implicated in a range of diverse biological
functions including the E3 ubiqutin-protein ligase activity that
determines the substrate specificity for ubiquitylation. Although zfp598
gene has found in many animals including human, mouse, dog, chicken
and frog, but there is not any reference about the biological function of
zfp598. After cloning and sequencing of zfp598 from zebrafish cDNA
library, our lab pretended to elucidate its biological function by in situ
hybridization and detected the differences between control and zfp598
morpholino injected notochord bended morphants by microarray.
Zfp598 was observed expression during head at 24, 48 and 72 hpf
larval with in situ hybridization. There was about 441 genes increased
expression over two fold and 421 genes decreased to half level in 72 hpf
morphant than control. The phenomenal of a lot genes influenced by
zfp598 knockdown indicated that zfp598 might play upstream gene to
regulated many genes during development.
