Identification and comparative analysis of the endosymbiosis - related molecules in cnidaria - dinoflagellate association / 刺絲胞動物與藻類共生相關分子之鑑定與分析

碩士 / 國立東華大學 / 海洋生物科技研究所 / 98 / Endosymbiosis in cnidaria-dinoflagellate association plays a critical role in regulating growth and productivity of corals and associated marine ecosystems. Cellular endosymbiosis with dinoflagellate, Symbiodinium spp. (i.e. the symbiont or “zooxanthellae” in generic name) within corals (i.e. the host) is initiated by the internalization of symbionts via phagocytic process into host gasterderm cells. However, the mechanism which the symbiont is able to reside inside of the host cell and then establish an obligatory and mutualistic association still remains ambiguous. For exploring the molecular mechanism of the endosymbiosis within cnidaria – dinoflagellate, synchrotron radiation-based infrared microspectroscopy(SR-IMS)and spatially - resolved nano - transmission X-ray microscopy (SR-nTXM) were used to study the molecular and morphological differences between symbiosome and free- living symbiodinium. It revealed that the chemical composition of symbiosome was different to free-living symbiodinium, especially the polysaccharide, lipid and nitrogen-related molecules. The 3D tomography images of symbiosome and free- living symbiodinium were also aquired by SR-nTXM. In the present study, the specificity of an antibody that preprared from symbiosome membrane protein were also analyzed. It demonstrated that the antibody not only recognized the protein with predicted molecular weight(MW) and isoelectric point(pI), but also recognized 3 proteins which with different molecular weight and isoelectric point. In future, it is needed to purify this antibody and identify it's target protein. In conclusion, the study provided the basic informations of endosymbiosis in molecular and morphological level.
