Influence of photoperiod and endosymbiosis on RuBisCO gene expression in Symbiodinium sp. / 光週期及胞內共生影響共生藻RuBisCO基因表現

碩士 / 國立東華大學 / 海洋生物科技研究所 / 98 / Symbiosis between corals and Symbiodinium sp. (zooxanthellae) is dependent upon the mutual exchange of nutrients. Symbiotic zooxanthellae could provide the photosynthetic nutrients to the coral host for exchanging the products from respiration metabolism from the host; and the host coral also protects and provides a habitat for the symbiotic algae zooxanthellae. In addition, the coral cells have been shown that regulating the photosynthetic activity of their symbionts. However, the mechanism of the mutual regulation between the host and its symbiont on the point view of photosynthesis remains unclear. Ribulose 1,5-bisphosphate carboxylase/oxygenase (RuBisCO) is the key enzyme of the carbon fixation reaction in the photosynthesis. The present study is to investigate the RuBisCO gene expression by semi-quantitive PCR and real time PCR techniques, and the RuBisCO protein expression using western blot analysis both in cultured and endosymbiotic Symbiodinium. The gene expression pattern has showed that fluctuations depend on circadian changes of light stimulation under light/dark photoperiod cycle in cultured Symbiodinium. On the other hand, endosymbiotic Symbiodinium in A. pulchella, the maximum RuBisCO transcript abundance occurred five hours after initiation of illumination, while the lowest RuBisCO transcript levels were measured at 14 hours. Results show that the fluctuating pattern of cultured Symbiodinium RuBisCO protein expression corresponded with diurnal change in light stimulation, and remained constant in endosymbiotic Symbiodinium. This study compared the RuBisCO gene and protein expression over diel cycles in both cultured and endosymbiotic Symbiodinium, and demonstrates the effect expression of RuBisCO under host-symbiont interaction status, and the correlating relationship of endosymbiosis and photosynthesis.
