碩士 / 國立高雄海洋科技大學 / 海洋生物技術研究所 / 98 / We have isolated 118 bacterial strains, including 23 Gram-positive strains and 95 Gram-negative strains, from a fry culture pond, a shrimp mariculture pond and a water area of saltpan. Phylogenetic analyses based on 16S rRNA gene sequences indicated that these strains belonged to 22 genera, such as Flectobacillus、Acinetobacter、Chitinilyticum、Pelomonas、Massilia、Enterobacter、Aeromonas、Andreprevotia、Bacillus、Klebsiella、Ideonella、Agrobacterium、Paracoccus、Flavobacterium、Chryseobacterium、Porphyrobacter、Pseudomonas、Exiguobacterium、Loktanella、Aquimarina、Neptuniibacter、Nautella and Marinobacter. Based on phylogentic analyses, strains GFA-11T, GFC-1T, an-8T and antisso-27 might represent four novel species. Therefore, they were subjected to identification and classification. And, investigating of algicidal activity in strain antisso-27 was performed because this isolate possessed potent ability to inhibit algal growth.
Strain GFA-11T, a Gram-negative rod bacterium, was isolated from a fry mariculture pond in Taoyuan county, Taiwan. Phylogenetic analyses based on 16S rRNA gene sequences showed that the closest neighbor strain is Flectobacillus major T (96.0%). The characterization of the genotype, phenotype and biochemical property demonstrated that strain GFA-11T represents a novel species of the genus Flectobacillus. We propose the name Flectobacillus roseus sp. nov., and the type strain is GFA-11T(=BCRC 17834T=LMG 24501T).

The characterization of the genotype, phenotype and biochemical property demonstrated that strain GFC-1T represents a novel species of the genus Andreprevotia .We propose the name Andreprevotia lacus sp. nov., and the type strain is GFC-1T(=BCRC17832T = LMG 24502T).
Strain an-8T, a Gram-negative rod bacterium, was isolated from a shrimp culture pond in Pingtung county, Taiwan. Phylogenetic analyses based on 16S rRNA gene sequences showed that the closest neighbor strain is Flavobacterium terrigenaT (95.1%). The characterization of the genotype, phenotype and biochemical property demonstrated that strain an-8T represents a novel species of the genus Flavobacterium .We propose the name Flavobacterium macrobrachii sp. nov., and the type strain is an-8T(=BCRC 17965T = LMG 25203T).
Strain antisso-27, a Gram- negative rod bacterium, was isolated from a water area of saltpan in Tainan county, Taiwan. Phylogenetic analyses based on 16S rRNA gene sequences showed that the closest neighbor strain is Aquimarina intermediaT (95.0%). The characterization of the genotype, phenotype and biochemical property demonstrated that strain antisso-27 represents a novel species of the genus Aquimarina. We propose the name Aquimarina salinaria sp. nov., and the type strain is antisso-27T(=BCRC 80080T= LMG 25375T).
Based on screening for algicidal activity, strain antisso-27 showed algicidal activity against several algae, including Cyanothece sp., Microcystis aeruginosa, Phormidium persicinum, Chlorella vulgaris and Nannochloropsis sp. The culture supernatant of strain antisso-27 possessed antimicrobial activity against Staphylococcus aureus. Strain antisso-27 was shown to synthesize an algicidal protein with a molecular mass of 195 kDa, and its isoelectric point is approximately 9.37 by isoelectric precipitation, SDS-PAGE, native-PAGE and activity assay. The results from Prussian blue agar test, Catalase effect assay and peroxidase-coupled assay suggested that the algicidal protein in strain antisso-27 was an L- amino acid oxidase. Algicidal activity was produced by generating hydrogen peroxide through this enzymatic activity of L- amino acid oxidase catalyzed L-leucine, L-isoleucine, L-methionine, L-valine. The algicidal activity occurred during the last stationary phase of the strain antisso-27 growth curve. Moreover, the algicidal protein was very stable at 4-70oC and showed a great resistance against acid-alkaline treatment (pH 4-11). The algicidal protein maintained full activity in solution at 25 oC at least 65 days.
