Study on the biological activities of Sargassum polycystum polysaccharides extractives / 匍枝馬尾藻多醣萃取物生物活性之探討

碩士 / 國立澎湖科技大學 / 海洋創意產業研究所 / 98 / It has been report that marine algae contain various biological active compounds. The polysaccharide especially from brown algae can enhance-immune activity and shows no toxicity. The aim of the present study was to investigate several biological properties of polysaccharide extracts from Sargassum polycystum include antimicrobial, antioxidant and antimutagenic effects.
Fraction C had better inhibitor effect on some gram-positive bacteria and gram-negative bacteria, including Bacillus subtilis, Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli and Morganella morganii. In the other hand, Fraction C also has antimicrobial activity against Zygosaccharomyces baili and Z. rouxi of fungi. The MIC values were 1.25 mg/ml and 2.5 mg/ml, and MBC values were 50 mg/ml and 20 mg/ml, respectively.
Fraction A(laminaran and fucoidan), Fraction B(fucoidan) and Fraction C were Sargassum polycystum crude polysaccharide extract. Fraction A was found to have the highest DPPH free radical scavenging ability(75.15%). In addition, Fraction C displayed the highest ferrous ion chelating ability and reducing power(2.72).
In mutagenicity and antimutagenicity assay, our results showed all polysaccharide extracts were no toxicity and mutagenicity with or without S9 mix. The antimutagenicity effect of Fraction A was the most significant against Salmonella typhimurium TA97(54.1 %) and TA100(53.0%) mutation induced by 4NQO. The antimutagenicity effect of Fraction B was also remarkable(63%)against Salmonella typhimurium TA100 mutation induced by B[a]P.
This study demonstrated that crude polysaccharides extracts from Sargassum polycystum have antimicrobial, antioxidant and antimutagenic ability. These polysaccharide extracts can be developed as natural preservatives or antioxidant in foods.
