Study on Norepinephrine Modulates the Immunity of Giant Freshwater Prawn, Macrobrachium rosenbergii / 正腎上腺素調控淡水長臂大蝦免疫能力之研究

碩士 / 國立屏東科技大學 / 水產養殖系所 / 98 / This study aimed to investigate the changes of the norepinephrine contents and immune factors in haemolymph when giant freshwater prawn, Macrobrachium resenbergii acclimated at 28℃, then sudden changed to high (34℃)and low(22℃)temperature. Using adrenoreceptor antagonists in vitro assay furher tested pathways of norepinephrine in modulating both immune factors and immune genes expression.
In M. resenbergii, the norepinephrine contents in haemolymph in high and low temperature were both higher than that of control (28℃) within 60 minutes, and in low temperature, the norepinephrine contents reached highest in all sampling time courses within 30 minutes. This study showed that norepinephrine contents were increased both in high and low temperature;immune factors showed temperature stress induced the release of norepinephrine brought phenoloxidase activity, superoxide anion and superoxide dismutase activity depressive effects within 120 minutes, but no significant effects observed in total haemocyte count.
The effects of norepinephrine in immune modulation showed that increase of phenoloxidase activity and phagocytotic activity primarily mediated through β1 adrenergic receptor, while respiratory burst primarily mediated through α1, β1 and β2 adrenergic receptors. In superoxide dismutase activity, adrengic effect primarily mediated through β2 adrenergic receptor. The norepinephrine modulated immune genes expression of proPO system showed adrenoreceptor antagonists treated haemocyte in proPO and LGBP primarily mediated through α1 and β1 adrenergic receptors, in α2-M and PE primarily mediated through α1 adrenergic receptor after 15 minutes incubation. When adrenoreceptor antagonists treated haemocyte for 30 minutes, all immune genes mediated only via α1 adrenergic receptor. That showed the adrenoreceptor antagonists of binding capacity at drenergic receptors decreased as the incubating time of haemocyte increased .
