碩士 / 國立臺灣海洋大學 / 生物科技研究所 / 98 / Zebrafish (Danio rerio) has been a prominent vertebrate model in a variety of biological disciplines. The animal model provides substantial information gathered from developmental and genetic research, together with completion of zebrafish genome, has placed zebrafish in an attractive position as a disease model. The tumor suppressor PTEN negatively regulates PI3K/AKT pathway that plays critical roles in cell growth, migration, and death. However, it’s less to know about the PI3K/AKT pathway involving in the early stage of molecular mechanisms during turmorigenesis. Here, we establish the regulating liver-specific compound transgenic (COMP) zebrafish systems which can individually and inducibly express several sets of genes at one time. By using those system, we perform over-expression NEDD4-a and co-inducible expression of K-ras led to decrease PTEN activity and company with increase of cell proliferation in COMP liver. The gene expression profiles and molecular mechanisms of cancerous phenotypes (tumorigenesis) will be analyzed in the near future. Eventually, the COMP lines will be applied to find novel therapeutic treatments and drug screen system for human liver disease.
