Cytochrome P450 1B1 (CYP1B1) is a monooxygenase which is expressed in eukaryotic endoplasmic reticulum (ER). CYP1B1 protein converts estradiol to 4-hydroxyestradiol by hydroxylation to reduce estrogen activity in animals. CYP1B1 proteins have been achieved catalytic activity in ER membrane, so this study was to use baculovirus expression system to express bioactive CYP1B1 protein. The western blot analysis results showed recombinant CYP1B1 protein was expressed in ER of sf9 cells. Due to the similar structure and function of estradiol and nonylphenol, nonylphenol was used to exam the bioactivity of recombinant CYP1B1 protein. Breast cancer cells, MCF-7, were treated with CYP1B1-nonylphenol metabilites, and theproliferation activity was measured by MTT assay. The results showed high concentration of nonylphenol after incubating with recombinant CYP1B1 inhibited the proliferation of MCF-7 cells. However, the metabolites from low concertration of nonylphenol increased the proliferation of MCF-7. In conclusion, the bioactive recombinant zebrafish CYP1B1 can be expressed in baculovirus expression system.
