Studies on the cosmetic bioactivities for astaxanthin and extracts of Glossogyne tenuifolia and Camellia oleifera / 蝦紅素及香茹、苦茶花萃取物之化粧品生物活性探討

碩士 / 嘉南藥理科技大學 / 化妝品科技研究所 / 99 / In this study, cytotoxic levels of astaxanthin, Glossogyne tenuifolia and Camellia oleifera flowers on 3T3 mouse fibroblasts were measured by MTT assay. Further, we investigated the inhibitory effects on activation of secreted matrix metalloproteinase-2 (MMP-2) and matrix metalloproteinase-9 (MMP-9) in culture medium using gelatin zymography. Collagen protection effect was researched subsequently using collagen assay kits. DPPH radicals scavenging activity, tyrosinase inhibition, agar disc diffusion method were performed for analyzing their antioxidant, whitening and antibacterial activity as well.
we observed that 150 μg/ml of astaxanthin aqueous solution which inhibited MMP-2, MMP-9 activities and reduced 89.08% of collagen decrement, was found 97.95±1.45% of high DPPH radicals scavenging activity. Both 10% of G. tenuifolia and 2% of C. oleifera flowers extract solution exhibited significant inhibitory effects on MMP-2, MMP-9 activities as well as complete protection effects for collagen. Moreover, they both manifested over 90% scavenging activity of DPPH radicals. In whitening and antibacterial experiment, these three subjects could not inhibit tyrosinase at all. Besides, astaxanthin aqueous solution and G. tenuifolia extract solution did not display obvious inhibition zones on Escherichia coli, Staphylococcus aureus and Pseudomonas aeruginosa. The extract solution of C. oleifera flowers appeared 12.3±0.5 mm of diameter of inhibition zones which is considered moderate activity on Staphylococcus aureus, but not shown antibacterial ability on the other two microorganisms.
Although these three subjects did not reveal powerful detections on whitening and antibacterial effects, they all possess strong antioxidant activities. Our results also demonstrate that astaxanthin aqueous solution, extracts from G. tenuifolia and C. oleifera flowers provide abilities to inhibit matrix metalloproteinases (MMPs), which is able to protect collagen fibers against degradation and leading to anti-ageing. This influence suggests that these three subjects may be considered effective ingredients for cosmetics application.
