Liver tissue engineering of the tidal culture test / 肝組織工程細胞之潮汐培養測試

碩士 / 輔英科技大學 / 生物科技系碩士班 / 99 / Liver cancer was the top three of cancers diseases in top ten causes of death in 2010 in Taiwan, announced by the Department of Health recently. Biological artificial liver support system, which can provide a good environment of growth and metabolism and exchange material, is becoming an effective way to treat liver failure. In this study, human hepatoma cell line (HepG2/C3A) was cultured by a tidal way. The growth and related liver functions using different substrates were evaluated. Initial results show that the attachment of cells on PC substrate was better than on PU substrate. Although the cell growth and albumin production by using both substrates in tide reactor were not as good as that we expected, the specific liver function of converting ammonia into urea nitrogen were retained. The feasibility of chitosan being a suitable substrate for artificial liver was also investigated in this research. The test was performed in static culture and the results show that the cell growth in pure chitosan was better than that in the different modified chitosan. Therefore, chitosan as a substrate of artificial liver is feasibility.
