Column Elution Chromatography Coupled with Supercritical Carbon Dioxide Antisolvent Precipitation of Carotenoids-rich Particulates from Dunaliella salina / 管柱流動層析結合超臨界二氧化碳抗溶沉澱杜莎藻富含類胡蘿蔔素粉粒體

碩士 / 國立中興大學 / 化學工程學系所 / 99 / This work investigated column elution chromatography of ultrasonic solvent extracts coupled with supercritical anti-solvent precipitation of carotenoids-containing particulates from microalgal Dunaliella Salina species. The extract contained carotenoids ranged from 61.3mg to 72.5mg using ultrasonic stirred ethyl ether or tetrahydrofuran (THF) extraction. When 6L ethyl alcohol was used in a reverse-phase column to elute 2g of the ethyl ether extract, the purity of trans-b-carotene attained 624.9mg/g, and the recovery was 84.6%. When 10L of ethyl alcohol employed for eluting the THF extract, the purity of trans-b-carotene increased to 823.6 mg/g with a recovery of 86.2%. Supercritical anti-solvent of ethyl ether solution did not enhance the purity of carotenoids, implied that carbon dioxide dissolves more carotenoids than chlorophylls into high-pressure solution. However, supercritical anti-solvent of THF solution at 160bar and 318K produced powdered particulates with purity of carotenoids above 90%. Subsequently, a center composited schemed response surface method designed supercritical anti-solvent precipitation of carotenoids-rich THF solution by changing pressure from 140bar to 160bar and time from 40min to 60min at feed flow rate of 0.2ml/min, CO2 flow rate of 15L/min and temperature of 318K was examined to find effects on the purity and recovery of trans-b-carotene. There experimental indeed produced micronized precipitates with mean particle size range from 3.5 micro meter to 19 micro meter and the purity of trans-b- carotene increased to 926.8mg/g with a recovery of 54%.
