Characteristics and purification of functional peptides from Tilapia skin protein hydrolysates / 吳郭魚皮蛋白水解物中機能性胜肽之特性探討與純化

碩士 / 國立高雄海洋科技大學 / 水產食品科學研究所 / 99 / Tilapia skin protein (TSP) was hydrolyzed by using proteinases with various hydrolysis times (1-11h). Radical scavenging activities (including 1,1-Diphenyl-2-picryl-hydrazyl (DPPH), hydroxyl, and superoxide anion radicals) of the TSP hydrolysates (TSPHs) were evaluated. In addition, angiotensin Ι converting enzyme (ACE) inhibitory activity and the protective effects on H2O2-induced DNA oxidative damage (DNA protective effect) of the TSPHs were investigated. The highest DPPH, superoxide anion, hydroxyl radical scavenging activities of TSPHs were 46.47, 34.20, and 45.49%, respectively. The highest DNA protective effect and ACE inhibitory activity of TSPHs were 54.82 and 87.06%, respectively. To understand the effect of molecular weight distribution of TSPHs on their functional properties, the TSPHs with the highest functional activity were fractionated using ultrafiltration. The radical scavenging, ACE inhibitory, and DNA protective activities of TSPH ultrafiltrates (TSPHUs) and FPLC-TSPH fractions were also evaluated. It indicated that TSPHUs and FPLC-TSPH fractions with molecular weight around 1 kDa exhibited excellent functional activities. It indicated that the highest DPPH, superoxide anion, hydroxyl radical scavenging activities of TSPHUs are 79.78, 46.79 and 25.13%, respectively. The FPLC-TSPH fractions were 30.39 (DPPH), 29.60 (superoxide), and 55.40 % (hydroxyl), respectively. The highest DNA protective effect and ACE inhibitory activity of TSPHUs were 106.34 and 50.18%, respectively. The highest DNA protective effect of FPLC-TSPH fractions were 79.19%. We further purified a potent antioxidative FPLC-TSPH fraction, PX5C, using RP-HPLC C18 column and identified its amino acid sequence. In summary, TSPH peptides may be developed as dietary supplements or functional food additives.
