碩士 / 國立臺灣海洋大學 / 水產養殖學系 / 99 / The aim on this thesis is studying the germ-cell marker genes on Euphyllia ancora. ie: VASA, VITELLOGENIN-LIKE, EGG PROTEIN-4 and ACETYLATED-ALPHA-TUBULIN. To know these marker genes have any pattern on different tissue on male or female colony and compare with higher evolutionary animals, coral regulatory mechsiam of germ-cell marker expression in year. Do GnRH and sex steroids effect germ-cell marker genes expression, We sampling from Februaty, 2010 to May, 2011 each month then using semi-quantitative PCR, western blotting and immunochemistry to analysis. we get VASA1, VASA2, VITELLOGENIN-LIKE and EGG PROTEIN-4 partial sequence from cDNA library and transcriptome sequencing then extending to get full lenth, making antibody. Treating coral with estradial, testosterone, LHRH-agonist and adipokinetic hormone to see genes change.
Results show that factors effect germ-cell marker genes chang including sex and seasons. Female colony express EGG PROTEIN-4 higher than male colony, spawning season higher non-spawning season in both sex. Three months after mass spawning, VITELLOGENIN-LIKE has lower expression. Egg doesn’t express EGG PROTEIN-4 when egg diameter smaller than 100 m from June to November, egg diameter between 200 m to 300 m only partial of egg express EGG PROTEIN-4, when egg diameter bigger than 300 m whole egg express EGG PROTEIN-4, however granule gland cell on mesentery filament and tentacle expresses EGG PROTEIN-4 every mont. spermatogonia expresses VASA in December. Spermatogonia expresses ACETYLATED-ALPHA-TUBULIN in October to spermatozoa in May, spermatid expresses is increasing in March, after spermatid become spermatozoa, major expressional position is flagellum. In treatment experiment, LHRH-agonist and AKH group have same pattern.
Coral spawning in similar timing each year. We can know which stage of gametes are and what are gametes doing. Is VITELLOGENIN and EGG PROTEIN-4 accumulated in the same time. Thus, combining with germ-cell development and factors which is effecting mass spawning is useful for studing physiology and reproduction on coral.
