Immunostimulatory effect of Ulva lactuca on white shrimp Litopenaeus vannamei and its resistance against Vibrio alginolyticus / 石蓴對白蝦免疫提昇效果以及促進其對於溶藻弧菌之抵抗

碩士 / 國立臺灣海洋大學 / 水產養殖學系 / 99 / In vitro tests were conducted by treatments with Ulva lactuca extract, powder, residue and filtrate to examine the induction of phenoloxidase (PO) activity and repiratory burst (RB) by the haemocytes of white shrimp Litopenaeus vannamei. The PO and RB of haemocytes that elicited by the 3 mg ml-1 Ulva extract, powder, residue and filtrate was significantly higher than those of 1 mg ml-1 Ulva extract, powder, residue and filtrate. No significant difference in PO activity was observed among the 1 mg ml-1 of Ulva extract, powder, residue, filtrate and β-glucan. The RB of haemocytes treated with 3 mg ml-1 extract was significantly higher than that treated with zymosan.
White shrimp L. vannamei which had been immersed in seawater containing the 200 mg L-1 Ulva extract, 500 mg L-1 powder and its residue and filtrate were challenged with Vibrio alginolyticus at 1 x 106 colony-forming units (cfu) shrimp-1, and then placed in seawater. Survival rates of shrimp immersed in seawater containing the Ulva extract, powder, residue and filtrate were significantly higher than that of control shrimp after 48-120 h.
In a separate experiment, the hyaline cell (HC) count, granular cell (GC, including semi-granular cell) count, total haemocyte count (THC), phenoloxidase (PO) activity, respiratory burst (RB), superoxide dismutase (SOD) activity, lysozyme activity, haemocyanin and haemolymph protein concentration were measured when shrimp were immersed in seawater containing the 200 mg L-1 Ulva extract, 500 mg L-1 powder and its residue and filtrate. Shrimp received extract had significantly higher immune parameters after 1 and 3 h. Shrimp that received the similar treatment and dosage via immersion had significantly increased compared with control shrimp in phagocytic activity and clearance efficiency to V. alginolyticus at 3 h.
It was concluded that white shrimp L. vannamei which had been immersed in seawater containing 200 mg L-1 Ulva extract, 500 mg L-1 powder and its residue and filtrate had enhanced innate immunity and increased resistance against V. alginolyticus infection.
