Blue Coloration and Body Color Regulation of Ornamental Fish / 觀賞魚藍色色素生成與調控機制

碩士 / 國立臺灣海洋大學 / 水產養殖學系 / 99 / The values of ornamental fish depend on size, appearance, color, and pattern. Shorten coloration time and color enhancing could bring more value to ornamental fish. The goals of present study is studying the mechanism of blue coloration and body color regulation of electric blue hap (Sciaenochromis fryeri). The partial sequence of purine nucleoside phosphorylase (PNPase) cDNA of electric blue hap was cloned by RT-PCR method. Partial sequence of PNPase gene contain of 109 bp in length, and the analysis of PNPase gene expression on electric blue hap skin revealed that adult fish has higher expression than junior fish. On the other hand, the junior fish treated with color enhancing formula feed has higher PNPase mRNA level in skin after 7 days treatment, this suggests that increasing gene expression of PNPase might increase the crystal purine plate in iridophores which can enhance the skin blue color. Skin melanophore number and blue area were increased after treated junior electric blue hap with color enhancing formula by 7 days. Treated with melatonin, the skin melanophore aggregated and lead to body color becam paler. After electric blue hap treated with color enhancing formula feed coupled with PTU after 7 days, skin melanosome contained melanophoer number and blue area were less than the fed color enhancing formula only. The body color was paler. No significant blue color difference by used extra pigment source, spirulina, Strobilanthes cusia and Lithospermum erythrorhizon, treatment on the blue body color of junior electric blue hap within 14 days. The application of color enhancing feed on ram cichlid (Mikrogeophagus ramirezi) showed deeper blue body color of fish than which treated with control after 7 days treatment. No difference in the color enhancing effect of All-plant protein source feed with color enhancing formula on junior electric blue hap and fish meal with color enhancing formula. No blue color enhancing difference was detacted between color enhancing formula conataining fish meal protein and soy bean substitution.
