Effect of sulfated polysaccharides on anticoagulant activity from Endarachne binghamiae / 小海帶含硫多醣對抗凝血活性之影響

The study was to investigate anticoagulant activities of crude sulfated polysaccharides (CSP) extracted from brown algae Endarachne binghamiae by hot water extracted (HWE) or acid hydrolysis (AH). CSP were extracted by hot water, the molecular weight are more than 3.5 kDa (HWE 3.5) or more than 5 kDa (HWE 5); by acid hydrolysis, the molecular weight are more than 5 kDa (AH 5). Total sugar of HWE 3.5, HWE 5, AH5 are 429.32, 427.33, 546.74 mg/g; sulfated groups are 165.90, 121.94, 113.82 mg/g. Ion exchange chromatography (IEC) with DEAE Sepharose CL-6B of the fractions of HWE 3.5 (IEC-HWE 3.5) and HWE 5 (IEC-HWE 5) showed much electric charge than IEC-AH 5. Gel permeation chromatography (GPC) with Sephacryl S-300 HR of the fractions of HWE 3.5 (GPC-HWE 3.5), HWE 5 (GPC-HWE 5) and AH 5 (GPC-AH 5) had the molecular weight between 155-275, 117-205 and 38-66 kDa. To prove the anticoagulant activities, we tested HWE 3.5, HWE 5, AH 5 and their frations of IEC and GPC by prothrombin time (PT) and activated partial thromboplastin time (APTT) with rabbit or human plasma. There were no effects on PT with rabbit plasma whether we used HWE 3.5, HWE 5, AH5 or their IEC and GPC fractions. However, HWE 3.5 showed best improved anticoagulant activities in APTT tests. The APTT of HWE 3.5, HWE 5, and AH5 were 300, 238 and 266 seconds when added 0.5 mg/mL into rabbit plasma individually. As for human plasma, HWE 3.5, HWE 5, AH5 and their IEC and GPC fractions all showed little effects in PT tests, therefore, they equips the potential of developing oral anticoagulant drugs. HWE 3.5 showed the promising anticoagulant activities in APTT tests, which proved the potential of developing injective anticoagulant drugs.
