Neuroprotective Effects and Mechanisms of Fucoxanthin Rich-Sargassum muticum Extract on Middle Cerebral Artery Occlusion Induced Focal Cerebral Ischemic Stroke In Rats / 富含藻黃素之墨角藻萃取物對大白鼠中大腦動脈阻塞誘發局部缺血性中風之神經保護作用與機制探討

碩士 / 國立臺灣海洋大學 / 食品科學系 / 99 / Ischemia strokes accounted for the majority of stroke cases (75 %). Cerebral ischemic stroke will cause further oxidative stress associated with the expansion of a variety of brain damage mechanisms. Therefore, there have been numerous studies using of antioxidant strategies to study ischemic stroke. Fucoxanthin has been shown to have good anti-oxidant and anti-inflammatory effects, and has been preliminarly confirmed the neuroprotective effect in hemorrhagic stroke. We use Sargassum muticum to extract fucoxanthin rich-brown algae extracts (FX-E) and determinate the content of antioxidants, which had 100 % radical scavenging activitiy of DPPH and ABTS on 3 g/mL and 5 mg/mL, then also contains polyphenols and flavonoids. To evaluate the neuroprotective effects of FX-E, we use CoCl2 to mimic hypoxic/ischemic conditions in neuron like cell PC12 and C6 glioma cell. Administration of FX-E can decrease cellular ROS, and increase in the survival of neuron cell. We also use middle cerebral artery occlusion induced focal cerebral ischemic stroke animal model to investigate the mechanism of neuroprotective effects of FX-E. However, we also can find reduction of inflammatory cytokine mRNA expression levels (TNF-α, iNOS, COX-2), and increased antioxidantion and detoxification genes mRNA expression level (Nrf2, HO-1), and also activity of antioxidant enzymes. According to above results, fucoxanthin might prossess neuroprotective possibility.
