effects of protein from bigeye tuna roe on the growth of human dermal fibroblasts / 大目鮪魚卵蛋白質對人類皮膚纖維母細胞生長之影響

碩士 / 國立臺灣海洋大學 / 食品科學系 / 99 / The purpose of this study was to examine the effect of proteins extracts from bigeye tuna (Thunnus obesus) roe on the cell proliferation and collagen production of human dermal fibroblast in vitro. Tuna roe used acetone defatting before or after phosphate buffered saline extraction. After then we used 80% saturation ammonium sulfate precipitation and frozen dry obtained bigeye tuna roe powder (AP and BTRP). AP and BTRP were yielded about 1.7% and 1.3%, but AP had higher protein content. SDS electrophoresis indicated the tuna roe principal protein M.W. was around 16 kDa. BTRP increased CCD-966SK (human skin fibroblast) proliferation up to 250% and led to stimulation of collagen production up to two folds at 48 hours. BTRP had better ability for fibroblasts proliferation than AP. AP and BTRP had slightly effect on HaCaT (keratinocytes) proliferation. AP and BTRP show good thermal stability even after 100 ℃. Therefore, our results preliminarily suggest that the key active components of bigeye tuna roe are low M. W. proteins/ peptides.
