碩士 / 國立臺灣海洋大學 / 機械與機電工程學系 / 99 / This thesis presents calcium alginate gel formation by the microelectrode of adjusting and controlling immediately, on applying to microarray cells and cells measurement, that can co-culture many kinds of cells are detected together and drug screening. It utilized by electrochemical method on ionizing the calcium ions from Calcium carbonate in calcium alginate solution, the solubilized Ca2+ can then bind alginate to induce calcium alginate gel formation.
According to the characteristic of hydrogenated amorphous silicon (a-Si:H), which impedance diminished after light illuminating. Not only used light to create real electrodes, but also combined optics setup with DMD projector to be the microelectrode of adjusting and controling immediately. Electrode patterns can be changed quickly and efficiently, and this method replaces the traditional microelectrode especially. Therefore it can control the positions and forms of calcium alginate gel formation what we want.
This research defines resolution of calcium alginate gel, the diameter of ligh spot is D0, and the diameter of calcium alginate gel is D, hence D/D0 define as resolution of calcium alginate gel. We learned various factors and relevance of resolution in experiment, long formation time and high voltage will cause bad resolution. Because high concentration of calcium alginate, it is difficult for ions to spread, cause the resolution is relatively good. High concentration of Calcium carbonate will cause poor resolution, but reaching the saturation concentration, the resolution is no obvious difference. The higher light frequency, the better resolution performance, but there is widely different between illuminative light frequency and without light frequency, finally we can grasp gel formation characteristic by controling the light frequency.
