The growth of osteoblast-like Saos-2 cells on cyanophycin membranes. / 類骨母細胞於藍藻蛋白薄膜上之生長探討

碩士 / 國立臺灣科技大學 / 化學工程系 / 99 / Cyanophycin granule polypeptide(CGP) is a non-ribosomal protein. It is synthesized in the majority of cyanobacteria as a temporary energy storage of nitrogen. The structure consists of a poly-aspartic acid backbone with arginine as a side chain. It is a kind of natural biological material.
In this study, glutaradehyde was used as a crosslinker for both soluble and insoluble cyanophycin. Genipin was used as a crosslinker for insoluble cyanophycin. Saos-2, an osteoblast-like cell line, has most of the properties of osteoblast cells. The cells were cultured on cyanophycin
membranes to examine the cell growth. MTT test was used to determine the cell viability. Alkaline phosphatase activity was analyzed by 4-nitrophenyl phosphate which could be converted to 4-nitrophenol with an absorbance at 405 nm.
The results showed that the cell viability on cyanophycin membranes were comparable to the control group and that the ALP activities of Saos-2 cells on the cyanophycin membranes were higher than the control group. In conclusions, cyanophycin showed a potential for Saos-2 cells proliferation.
