Physicochemical Properties of the Cross-Linking Reactions Between Chitosan /Alginate Blend Membrane and Genipin / 以天然交聯劑Genipin交聯幾丁聚醣/海藻酸鈉材料的物理化學性質研究

碩士 / 亞東技術學院 / 纖維與材料應用產業研發碩士專班 / 99 / In this study non-toxic Genipin cross-linker with Chitosan (C), Alginate (A),Chitosan / Alginate (C1A1, C1A3) under the conditions of cross-linking at room temperature for 24 hours, and changing conditions of temperature and time of preparation of GCC, GCA and GCC1A1, GCC1A3 film. Crosslinked film using infrared spectroscopy bonding reaction. And thermal properties, mechanical properties, dynamic mechanical properties, absorbing and desorbing, antibacterial properties and biocompatibility, and physicochemical properties were studied.
Infrared spectroscopy shows that in the GCCA film on the FT-IR found in the 1261 cm-1 ; 1738 cm-1 found a new characteristic absorption peak, genipin and chitosan by the reaction of the six-ring diene three nitrogen amine absorption peaks / genipin and the reaction of sodium alginate ether characteristic absorption peaks; genipin 2 or 9 carbon of -OH/-CH2OH and alginate on the 6th carbon of the acid-base reaction of dehydration ester bond to generate the characteristic absorption peak. Thermal properties results show that chitosan glass transition temperature of 155 ℃, the glass transition temperature of sodium alginate as 108 ℃, blended C1A1 film glass transition temperature of 112 ℃, 0.2wt% GCC1A1 glass transition temperature of 129 ℃ the film in this ratio under compatibility blended addition, cross-linked film glass transition temperature increases. Mechanical properties showed that cross-linked GCC1A1 film processing temperature 60 ℃, the maximum processing time of 60 minute tensile strength 7.18 kg / cm2. Crosslinked GCC1A1 film tensile strength of about 3.0 the film blends C1A1. Dynamic mechanical properties show that cross-linked film storage modulus values lower than those not cross-linked structure, but only a slight decline. All material damping small, 200 ℃ temperature to stabilize the film. SEM observation showed that cross-linked GCC1A1 film cross-section shape a clean, smooth and flat, slice layer will increase as the crosslinker concentration increased film thickness. Crosslinked GCC1A3 film cross-section shape is found by a few layers between the numerous small segment link, the big sheet of layers disappear, high-magnification cross-section diagram found on numerous spherical or block the surface sediment layers together to form a clear interface stacked map. Absorption release show, blended with the crosslinked GCC1A1 C1A1 film moisture 2.0wt% tranexamic acid after 15 minutes, can be successful through 100 ~ 200nmPE material of the artificial skin and released. Sterilization rate show, during and after the sterilization of chitosan film was 100%, blended C1A1, C1A3 film sterilization was only a slight decrease, 98.07 and 95.36%. Crosslinking GCC1A1, GCC1A3 film sterilization rate is insignificant, the range is 82.69-89.75% for sterilization. Observation that cell types, CA and GCCA film compared with the positive control group, CA with GCCA film does not inhibit cell growth, so the film is not toxic to cells.
