碩士 / 靜宜大學 / 食品營養研究所 / 99 / Spirulina platensis contains abundant of protein, vitamin B12 and C-phycocyanin (C-PC). It was demonstrated that Spirulina platensis and C-PC have potently physiological properties, such as excellent antioxidant ability, neuron protection, virus inhibition and lessen the inflammation. However, theeffects of Spirulina platensis and C-PC on the anti-inflammatory activity of microglial cells is unclear. In this in vitro study, we aimed to evaluate the anti-inflammatory and antioxidant effects of Spirulina platensis water extracts and C-PC for lipopolysaccharide (LPS)-stimulated inflammation reaction with BV-2 cells. The experiment was devided into control, LPS or LPS added different doses of Spirulina platensis extracts and C-PC groups. First, we determined the influence of Spirulina platensis extracts and C-PC in BV-2 cell viability and cytotoxicity, then examined the oxidative damage status by reactive oxygen species (ROS) and nitric oxide (NO) generation. At the same time, we also evaluated the production of Prostaglandins E2 (PGE2), and gene expressions of inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), tumor necrosis factor-α (TNF-α), interleukine-1β (IL-1β) and interleukine-6 (IL-6). The results showed that Spirulina platensis extracts and C-PC could significantly decrease the LPS-induced cytotoxicity, and reduce the ROS, NO and PGE2 productions. Spirulina platensis extracts and C-PC also significantly diminished the TNF-α and IL-6 mRNA expressions, whereas they tend to lower the gene expressions of iNOS, COX-2and IL-1β without significantly difference. In summary, Spirulina platensis extracts and C-PC showed good antioxidant ability and could inhibit the expression of inflammatory factors in actived microglial cells.
