Preventative effect of algal extract against arecoline-induced cell growth inhibition through the apoptosis, ROS, and NO pathway / 海藻提取物對檳榔鹼誘導的細胞生長抑制是經由細胞凋亡、活性氧和一氧化氮的途徑

碩士 / 高雄醫學大學 / 天然藥物研究所 / 100 / In recent years, because of people''s lifestyles and eating habits, the frequence of various types of cancer becomes higher and higher. Therefore, a new trend of anti-oxidants will be able to deepen the discussion, research and development. Extracted from the algal extract, the abstracts have potential anti-oxidants structure separation. Arecoline, a major alkaloid of the areca nut, causes cell toxicity because of enhancement of oxidative stress and suppression of the anti-oxidant protective system. Algal extract of Gracilaria spp. is a well-known source of antioxidants and it has been proved to have a potential biological medicinal effectiveness such as anticarcinogenesis . The algal extract of hot extract has potential anti-oxidantion properties. Accordingly, the hypothesis that the possible protective effects of hot extract were against arecoline was proposed. In this study, using different concentration, 0.1, 0.125, 0.25, 0.35, 0.5 and 0.6 mg / ml, of hot extract and arecoline of various concentrations of 0.1, 0.25, 0.5, 0.75 mM and Ca9-22 oral cancer cells cultured for 24 hours, the impact of cell differentiation. To examine this hypothesis, the MTS assay of survival analysis, cell cycle analysis, apoptosis, reactive oxygen species (ROS) and Nitric Oxide (NO) were planned to finish in my thesis. Currently, the arecoline inhibit cell from growing. Besides ROS increase and NO increase in oral cancer cell line Ca9-22 were both caused the by arecoline. Furthermore, ROS increase and NO increase in oral cancer cell line Ca9-22 can mend through the hot extract. Once all expected work is done, it will provide a clear statement for the possible protection that hot extract is against arecoline-induced DNA damaging effects in oral cancer cells. Studies have confirmed the hot extract can inhibit the oxidative damage caused by arecoline on cell.
