Cloning of Kennedy pathway genes for enhanced storage lipid production in transgenic microalgae / 探討Kennedy pathway基因表現對微藻油脂含量之影響

碩士 / 明志科技大學 / 生化工程研究所 / 100 / In order to develop the microalgae strains which produce highly efficient and critical enzyme during lipid synthesis and metabolism, this study utilizes the genetic engineering approach to designs a novel expression vector for higher efficiency of oil production in Microalgae. in this research, we develop a series of expression vectors, four promoters (PCaMV35s, PRBCs, PAMT, PNR) were selected for driving the foreign genes (GPAT, LPAAT, PAP, DGAT) from Chlamydomonas and transforming into Micractinium sp.. In the experimental results, the expression level of the applied promoters was PAMT>PRBCs>PCaMV35s>PNR and the expression level of PCaMV35s to drive the foreign genes was PAP>LPAAT>DGAT>GPAT. In addition, the gene LPAAT-PAP-DGAT had the best performance of lipid content of microalgae with FAME and crude lipid increased by 48.9% and 68%, respectively. In conclusion, this study effectively used genetic engineering to increase the lipid content of microalgae and provided a novel method for microalgae biofuel production.
