Enhance Microalgae Lipid by Using UV-Mutation Strategies / 以紫外線突變策略提升微藻油脂含量之研究

碩士 / 明志科技大學 / 生化工程研究所 / 100 / In this study, algae mutants were generated by UV radiation with various exposure time and distance. The starch contents of algae strains were examined by iodine vapor and starch analysis kit while the lipid contents were determined by gas chromatography analysis. We attempted to improve the efficiency of lipid production in microalgae.
The result of UV-induced mutagenesis showed that the 40 seconds exposure time would completely inhibit the growth of microalgae, about 658 algae strains were obtained under various UV exposure conditions; 16 strains out of 96 candidates were randomly selected and subjected to subsequent starch quantification, colorimetric method quantification and GC analysis. The results showed 15 out of 16 strains possess lower starch contents (with a 31.4% decrease at most) compared to those of wild-type, whereas the lipid contents increased by about 84.3%.
16 mutants were cultured in tube photobioreactor and showed that their excellent crude lipid and lipid productivity. The efficiency of lipid productivity of 2-week culturing is much better than 1-week, M3, M5, M6, M10 and M12 mutants showed the most promoted lipid producing rate to 130.0, 106.1, 105.3, 115.0 and 107.1 mg/L/d respectively. under 2-week culturing, M3 mutant showed the best lipid producing rate, which is 1.99 times than wild type, besides, its biomass productivity is higher than wilt type by 12% in the second week. Therefore M3 is an extremely potential strain for lipid producing.
