碩士 / 國立中興大學 / 化學系所 / 100 / In this study, a simple and sensitive method using modified magnetic nanoparticles (C8@Fe3O4) extraction combined with liquid chromatography-tandem mass spectrometry (LC-MS/MS) for determination of trace tetrodotoxin in sea food was evaluated. Tandem mass spectrometry with positive electrospray ionization mode was used to detect tetrodotoxin. Tetrodotoxin was extracted from sea food by using 1 mL acetic acid/ethanol/water solution (5/20/75, v/v/v), then extracted by 250 mg of C8@Fe3O4 nanoparticles for 5 minutes. After extraction, 500μL of 0.5% acetic acid was used to desorb the tetrodotoxin from C8@Fe3O4 nanoparticles, then the extractant was analyzed by LC-MS/MS. The linear range of proposed method was 5-1000 ng/g with coefficient of determination at 0.9946. The limit of detection and limit of quantification were 1.4 and 4.8 ng/g, respectively. The intra-day and inter-day precision expressed by relative standard deviation were less than 3.6 and 8.9 %, respectively. The feasibility of applying the proposed method to analyze trace tetrodotoxin in commercial sea food was also examined. The results showed no tetrodotoxin was detected in the commercial sea food.
