Due to the limitation of fossil fuel resources, oil prices have being gradually increasing. Besides, combustion of fossil fuels will increase carbon dioxide in the atmosphere, resulting in the greenhouse effect. Therefore, it has become an urgent issue which everyone concern. Until now, many countries have started to concern about the development of biofuels since it has great potential to use macroalgae as biofuels. In this study, Gracilaria tenuisptitata was used to be the alternative raw materials of biodiesel, which respectively cultured in five different temperatures, 10, 15, 20, 25 and 30 ° C, three kinds of different photoperiodism, L: D = 12:12 h, 24:0 h and 0:24 h, and the cultivated medium lacking N, Fe and Cl for six days, then the freeze drying mode was used on the subject, and then its lipid was extracted through the soxhlet extraction, so as to measure the lipid content of macroalgae. The results show that Gracilaria tenuisptitata lipid accumulation significantly affected by different temperature (p <0.001) and the highest lipid accumulation occurs at 20 ° C and it’s lipid content is 15.62 mg/g DW. The different photoperiodisms significantly affect the lipid accumulation of this seaweed (p<0.001). In the absence of light, the lipid content was significantly reduced (5.14 mg/g DW), and in the absence of nitrogen sources, the lipid accumulation in this macroalga increased (18.5 mg/g DW).
