Molecular cloning and characterization of superoxide dismutase from the scleractina corals, Euphyllia glabrescens and Tubastraea aurea / 束形真葉珊瑚與圓管星珊瑚超氧歧化酶基因選殖之研究

碩士 / 國立東華大學 / 海洋生物科技研究所 / 100 / The purpose of this study was cloned different types of superoxide dismutase (SOD) genes from Euphyllia glabrescens, symbiodium and Tubastraea aurea. We use polymerase chain reaction (PCR) and rapid amplification of cDNA ends (RACE) for gene cloning,were isolated copper-zinc SOD (CuZnSOD) and mitochondrial manganese SOD (mtMnSOD) of full-lengths from E. glabrescens, CuZnSOD and mtMnSOD of the partial sequence from T. aurea, manganese SOD (MnSOD) and iron SOD (FeSOD) of the partial sequence from symbiodium. The E. glabrescens CuZnSOD gene length of 896 bp and open reading frames encoded 206 amino acids. Deduced peptide was molecular mass of 22kDa; and we found that the sea anemone (Anemonia viridis, AAS98800) identity with 52%. The E. glabrescens mtMnSOD gene length of 1096 bp and open reading frames encoded 223 amino acids; and contains two potential glycosylation binding NHS and NLS. Deduced peptide was molecular mass of 24kDa; the coral (Stylophila pistillata, AAX99423) identity with 86%.
