Preparation of collagen peptides with calcium binding activity from milkfish ( Chanos chanos ) scale / 虱目魚魚鱗膠原蛋白胜肽的製備與鈣結合能力探討

碩士 / 國立高雄海洋科技大學 / 水產食品科學研究所 / 100 / The objective of this study is to establish the process conditions for preparing collagen peptides with calcium-binding (Ca-B) activity from milkfish scales. The scales of milkfish was washed, dried, and milled to obtain a powder sample. Collagen peptides with Ca-B activity were prepared from milkfish scales powder treated with heating, acidic treatment and commercial proteinase(alcalase, papain, protase, protamex, flavourzyme). The optimal conditions in heat, acidic and enzymatic treatment were investigated and compared for their Ca-B activity. The hydrolysates from heat and acidic treatment did not have high Ca-B activity. Higher Ca-B activity was obtained from milkfish scales treated with alcalase, papain and flavourzyme. The milkfish scales were then treated with two enzymes system, including alcalase plus flavourzyme and papain plus flavourzyme. Those from alcalase plus flavourzyme showed higher Ca-B activity. The optimal conditions in two enzymes system were listed as follows: enzyme activity, 3000 U/mL for alcalase, 9000 U / mL for flavourzyme; pH 7.5; temperature 50 °C; reaction time 2 h ; 6 % for concentration of milkfish. The hydrolysates from the above two enzymes treatment were found to have similar Ca-B activity when compared with commercial casein phosphopeptide ( CPP ). High Ca-B activity was still found in the above hydrolysates after treated with artificial gastrointestinal liquid. The hydrolysates form milkfish scales were then applied in juice product, good overall acceptability was found according to sensory evaluation.
