Effects of Dietary Supplementation of Carotenoids on the Pigmentation, Antioxidation capacity, Non-specific immune responses and Disease resistance of the Red Tilapia(Oreochromis sp.) / 飼糧類胡蘿蔔素對紅色吳郭魚(Oreochromis sp.)呈色、抗氧化能力、非特異性免疫反應及抗病能力之影響

碩士 / 國立高雄海洋科技大學 / 水產養殖研究所 / 100 / This study aimed to determine whether dietary carotenoid supplements could affect the pigmentation, antioxidant capacity, non-specific immune responses and disease resistance of the red Tilapia(Oreochomis sp.). Natural carotenoids from astaxanthin containing yeast Phaffia rhodozyma (P), alga Haematococcus pluvialis (H) and a synthetic astaxanthin carophyll pink (A), β-carotene (B) were supplemented in formulated diets at two concentration, 80 and 160 mg/kg, resulting in nine pigmented diet P8, P16, H8, H16, A8, A16, B8 and B16. Formulated diet without carotenoid supplementation served as a control (C). The different diets were fed to red Tilapia(Oreochomis sp.), for 12 weeks. Serum total antioxidant status (TAS), serum antioxidant enzyme[superoxide dismutase (SOD), glutathione peroxidases (GPx)]and serum transaminases[alanine aminotransferase (ALT), aspartate aminotransferase (ALT)]were chosen as indices of fish antioxidant capacity or stress resistance. Innate immune (phagocytic activity, respiratory burst, lysozyme activity, superoxide dismutase and glutathione peroxidase activity) were evaluated. No different in weight gain and survival rate were found among all fish. There were higher pigment content in pigment treatment than control. The pigmentation of astaxanthin in the fish of natural astaxanthin-fed, and artificial astaxanthin-fed fish were higher than those of fish fed without astaxanthin. There were higher pigment concentration in A16, H16 treatment than those of fish fed other pigment treatment and the control diet. The pigment treatments had higher TAS and GPx than the control fish. Dietary carotenoid reduce SOD, ALT, and AST activities. Pigment treated fish showed significantly lower respiratory burst and SOD, higher GPx, phagocytosis index and lysozyme activity compared with the control groups. In the challenge group, pigment treatments had significantly higher survival rates than control. The survival rates 168h after challenge were 82.03 % and 18.75 % for pigment groups and control, respectively. There were significantly differences in survival rate were found between pigment treatments and control fish afterwards.
