Effect of cortisol on calcium uptake in tilapia (Oreochromis mossambicus) / 皮質醇調節吳郭魚鈣離子吸收作用

碩士 / 國立臺南大學 / 生物科技學系碩士班 / 100 / Calcium is an essential element for all animals. In fish, gill is a main organ on calcium uptake. Cortisol which is a corticosteroid was reported was proposed to show hypercalcemic effects in teleostean fish. Corticosteroid involved in regulating the osmolality and ion levels, body fluids, energy metabolism, respiration, and immune reactions. Cortisol was previously found to regulate Ca2+ uptake in fish, but the mechanism is still unclear. Tilapia has good tolerance on salinity and well study on ion regulation. We use tilapia be a model here. In this study, we want to know the molecular mechanism of cortisol on calcium uptake in tilapia.
In low calcium fresh water, the expression of epithelial calcium channel (ecac) and the mineralocorticoid receptor (mr) was increased. Exogenous cortisol increased whole body calcium content and the expression of epithelial calcium channel (ecac), but the effect did not show on other calcium transporters or the mineralocorticoid receptor (mr) and the glucocorticoid receptor (gr). Exogenous cortisol with GR and MR antagonist decreased the expression of epithelial calcium channel (ecac). In conclusion, cortisol stimulates epithelial calcium channel (ecac) expression to enhance calcium uptake functions, and this control pathway is suggested to be mediated both the GR and MR.
