碩士 / 國立臺灣海洋大學 / 水產養殖學系 / 100 / White shrimp Litopenaeus vannamei juveniles (0.48 ± 0.05 g) which had been reared at different salinity levels of 2.5, 5, 15, 25, and 35‰ for 24 weeks were examined for the survival, growth, immune parameters, resistance against Vibrio alginolyticus and WSSV, haemocyte life span, and expressions of lipopolysaccharide and β-glucan binding protein (LGBP), peroxinectin (PX) integrin β, and α2-macroglobulin (α2- M). All shrimp survived during the first 9 weeks. The survival rate of shrimp reared at 2.5 and 5‰ was much lower (30%) compared to shrimp reared at 15, 25 and 35‰ (76~86%) after 24 weeks. Shrimp reared at 25% grow faster and better compared to other salinity. Shrimp reared at 2.5 and 5‰ showed lower hyaline cells (HC), granular cells (GC), phenoloxidase activity (PO) activity, respiratory burst (RB), superoxide dismutase (SOD), and lysozyme activity, but showed longer life span of haemocyte, and higher expressions of LGBP, PX, integrin β, and α2-M compared to shrimp reared at 15, 25 and 35‰. Shrimp that were reared in different salinity for 24 weeks were challenged with V. alginolyticus (6.5 x 106 cfu shrimp-1), and WSSV (1 x 106 copies shrimp-1) and then released its respective seawater. Over 96~144 h, the cumulative mortalities of shrimp reared at 2.5 and 5‰ were significantly higher than those of shrimp reared at 15, 25, and 35‰. It was concluded that white shrimp L. vannamei juveniles following long-term exposure to 2.5 and 5‰ exhibited decreased resistance against pathogen by the reduction of immune parameters. Increases of haemocyte life span and gene expressions of LGBP, integrin β, PX and α2-M indicated that shrimp showed an extra energy to modulate innate immune system of proPO system in preventing further perturbation in low salinity levels.
