Studies on a 24-hours choline metabolism of hybrid tilapia(Oreochromis aureus x O. niloticus) with modified cannulated urine collector / 使用改良型尿液收集器於吳郭魚24小時膽鹼代謝之研究

碩士 / 國立臺灣海洋大學 / 水產養殖學系 / 100 / The purpose of this study was to investigate the choline metabolism in hybrid tilapia(O.aureus (♀) × O. niloticus (♂)) by two input methods with urine collection, in order to determine the absorption and utilization through the 24 hour course of choline concentration variation in the blood and urine.
There were two experiments the forced feed 0.3% body wet weight ration level experiments and injection experiments. The forced feed groups of fish were feed orally adminstrated diet choline level 0, 200, 400, 600, 800 and 1600 ppm. In the injection experiment, fish were injected choline solution at 0, 10, 20, 50 and 100ppm level with 5ml into the vein of tail. The urine and blood samples were collected at 2, 4, 6, 8, 10, 12, 16, 20, 24 hours, after oral or injection administration of choline. Muscle、kidney、heart and liver samples were obtained at the 24th hour in the end of the experiment.
It was observed that the plasma concentrations of the whole treatments were all peaked at the 6th-8th hour and urine concentration peaked at the 12th hour after oral administrations of diets. The plasma choline peaked at the 2nd hours ,while that for choline was at the 8th hour after injection. The delay of 4-6 hours of the plasma choline concentration peak and the delay of 4 hours of the urine choline concentration peak in the oral administration group of fish than the respective two peaks in the injection fish group revealed.
Inulin clearance test showed the glomerular filtration rate (GFR) was 16.87~22.81 (ml / min) at the 120min after injection. The total choline excretion increased with increasing injection choline level.
The tilapia choline recovery rate at 2minutes to 2hours is between 98.23%~99.96%,the low concentration is higer than high concentration.
The ADME (absorption, distribution, metabolism, excretion, experiment shows the unknown will increase by choline additional,and forced feed groups’ unknown will higher than injection groups.
