Study on the species identification, cultivation, and contents and compositions of the mucilage of Spirulina platensis / 布藍特螺旋藻之品種鑑定、培養及其膠質的含量及成分分析之研究

博士 / 國立臺灣海洋大學 / 水產養殖學系 / 100 / This study investigated the cultivation conditions of Spirulina platensis, conducted species analysis on a number of samples, and analyzed the quantity and composition of its mucilage components. A total of ten fresh cultures and six commercial powders were collected in Taiwan. The samples were identified as S. platensis and S. maxima through analysis of the internal transcribed spacer (ITS) sequences of the samples.
The results of this study indicate that high intensity light (1320 μmol m-2 s-1) can increase the biomass of S. platensis and the optimal temperature of the growth environment is 28 °C. High concentrations of NaCl (0.75 M), however, reduce the biomass of this microalgae. Three types of light sources were employed to cultivate S. platensis: natural light, fluorescent lamps, and white cold DDH GaAlAs LEDs. No significant differences were observed when using the LEDs, indicating their potential application as an alternative to fluorescent lamps capable of considerably reducing costs and the space required for cultivation.
One of the objectives of this study was to analyze the composition of the mucilage in S. platensis, due to the high pharmaceutical value of the polysaccharide. The content of mucilage extracted from fresh cultures of S. platensis was found to range from 3.70 ± 0.52 to 27.31 ± 2.83%. The composition of the polysaccharide included 1.94 % of sulfate radical ions and four kinds of metal ions: sodium (1.88 %), potassium (0.27 %), magnesium (0.40 %), and calcium (0.79 %).
The polysaccharide obtained from fresh cultures of S. platensis were divided into two fractions using gel filtration chromatography: high molecular weight greater than 4.73 × 104 MW, and low molecular weight ranging between 5.90 × 103 and 1.18 × 104 MW. The above two fractions and unfractioned polysaccharide were subjected to acidolysis followed by high-performance liquid chromatography (HPLC) using an H form column. Seven different kinds of components were detected, including fucose, galactose, glucose, maltose, rhamnose, erythritol, and glycerol. These hydrolysates were further subjected to HPLC with a Ca form column, resulting in five types of components: arabinose, galactose, glucose, rhamnose, and mannitol. The unfractioned and high molecular weight of polysaccharide presented a phenomenon of particular interest, in which the hydrolysates analyzed by the Pb form column revealed six types of components: glucose, mannose, galactose, rhamnose, erythritol, and mannitol. In contrast, low molecular weight of polysaccharide resulted in only five types of components in the analysis with a Pb form column: mannose, rhamnose, glucose, galactose and erythritol.
Results from analyses using three types of ion columns revealed ten components in the unfractioned polysaccharide which include six monosaccharides, arabinose, fucose, galactose, glucose, mannose and rhamnose; one disaccharide, maltose; and three sugar alcohols, erythritol, mannitol and glycerol. Across the polysaccharide samples, the high molecular weight of polysaccharide exhibited higher contents in five components than low molecular weight of polysaccharide, including galactose, glucose, rhamnose, glycerol and mannitol. In comparison, the low molecular weight of polysaccharide contained more arabinose, fucose, maltose, mannose and erythritol than the high polysaccharide. This study employed ITS to identify the species of Spirulina currently being cultivated and sold on the market in Taiwan. This study developed a novel approach to the cultivation of Spirulina using LEDs to conserve energy and provided a quick, repeatable, and accurate analysis method for the quantification of polysaccharide components in S. platensis.
