The Effect of Chinese Herbal Compound (Xiao-Chai-Hu-Tang) on Non-specific Immune Responses of Orange-spotted Grouper (Epinephelus coioides) / 中藥複方小柴胡湯對點帶石斑魚非特異性免疫反應之影響

碩士 / 國立臺灣海洋大學 / 水產養殖學系 / 100 / This study uses in vitro and in vivo experiment to investigate the effect of Chinese herbal compound (Xiao-Chai-Hu-Tang) on growth and non-specific immune parameters of grouper (Epinephelus coioides). In vitro experiment in this study uses various doses (0、0.1、0.5、1、2、4、8、10 mg/ml) of Xiao-Chai-Hu-Tang extract for the in vitro culture of orange-spotted grouper leukocytes. The result showed that leukocytes of grouper culture with over 1 mg/ml of Xiao-Chai-Hu-Tang had significantly (p<0.05) enhanced the production ability of superoxide anion. In vivo experiment was conducted by adding Xiao-Chai-Hu-Tang at different dosage (0 %, 0.5 %, 1.0 %, 2.0 %) in the diets and was administered to the grouper for 30 days. Analysis of the growth and the effect non-specific immune parameters of grouper. After fed 30 day was intraperitoneal injection of Vibrio alginolyticus, and observed and recorded the current deposit situation after challenge. The result for immune parameters showed that the production ability of superoxide anion, reactive oxygen species,phagocytic ratio and phagocytic index, and lysozyme enzyme activity of plasma. Both increased non-specific effect of immune parameters. The results showed that the growth in continuous feeding of grouper add Xiao-chai-hu-tang feed 30 days, it’s no significant effect on its growth. The result for immune parameters showed that the production ability of superoxide anion, reactive oxygen species,phagocytic ratio and phagocytic index, and lysozyme enzyme activity of plasma, both increased non-specific effect of immune parameters. Challenge experiments showed that dietary Xiao-chai-hu-tang can effectively enhance grouper by V. alginolyticus challenge the current deposit rate. Comprehensive results of this study found that dietary herbal compound Xiao Chai Hu Tang, grouper grow to the point with no negative impact, can effectively enhance its non-specific immune response and against Vibrio alginolyticus infection.
