碩士 / 國立臺灣海洋大學 / 食品科學系 / 100 / Both clam (Corbicula fluminea) and Chlorella sorokiniana have antioxidative and hypolipidemic activities. The purpose of this study was to elucidate the combination effect of freshwater clam and Chlorella sorokiniana on antioxidative, prolongation of LDL oxidation and preventive of atherosclerosis using tilapia as an alternative animal model. Freshwater clam and Chlorella sorokiniana were extracted with hexane, ethyl acetate, ethanol and water. The in vitro model, showed that Chlorella hexane extract (Ch) at 1mg/ml has the strongest inhibitory effect against 5-, 12-, 15-lipoxygenase (LOXs), protecting Cu2+-induced oxidation for human and tilapia low-density lipoprotein (LDL). The inhibitory effect of hot-water extract of freshwater clam (FC-HW) was better than other solvent extracts of clam. Combined Ch and FC-HW in 1 : 1 (w/w) ratio, the LDL oxidation was prolonged better than FC-HW alone, but with no synergistic effect.
Tilapia were fed 1% Ch and 1% FC-HW for 8 weeks, significantly reduced total cholesterol and LDL concentration in plasma. Tilapia fed with Ch or FC-HW increased total antioxidant capacity (TAC) in plasma. Ch increased TAC of LDL, while FC-HW increased TAC of the aqueous phase of plasma.
Tilapia LDL oxidation was used as an ex vivo model. Tilapia fed 1% Ch had the fastest oxidative rates, while the combination extracts prolonged oxidation rates of LDL and lowered the maximum oxidative state .
Analyzed the lipid-soluble composition in plasma, the content of α-tocopherol showed no significant difference among different group. Tilapia fed 1% Ch had significantly higher content of lutein in plasma and LDL.
Using tilapia as an in vivo model showed that water-soluble and lipid-soluble antioxidants have synergistic effect to prolongate LDL oxidation.
