碩士 / 國立臺灣海洋大學 / 海洋生物研究所 / 100 / Lulworthia (Lulworthiaceae, Lulworthiales) is a predominantly marine genus belonging to the phylum Ascomycota with 11 species. The main taxonomic problems of the genus include: (1) ascospores of Lulworthia species are similar in morphology and the dimension of ascospores and ascospore appendages overlaps between different species, and (2) other morphological characters in Lulworthia are variable, including position of the ascomata in the substrate and presence or absence of neck. As a result, different authorities hold different opinions on the morphological characters used to delimit different species. For the last few years, many Lulworthia species have been collected from various mangrove forests and rocky shores in Taiwan and Denmark. A phylogenetic analysis of the partial 18S and 28S rRNA genes was initiated to investigate the phylogeny of these fungi. Morphology of these fungi was also examined to determine if morphological features of these fungi were concordant with their phylogeny. Maximum parsimony analysis of the sequences suggested that the genus Lulworthia is not monophyletic. A new genus should be established for Lulworthia grandispora due to the distant phylogenetic relationship with the type species, L. fucicola. Paraffin sections of the L. grandispora isolates consistently showed the presence of several layers of thick-walled cells at the basal part of the necks, a character not reported in other Lulworthia spp., except a similar structure in Rostrupiella. Lulworthia sp. JK1009 from Denmark grouped with a clade comprising several isolates of Lulworthia floridana, and Lulworthia sp. NTOU3706 clustered with Moromyces varia, suggesting a possible teleomorph/anamorph relationship but no conidia were observed from agar cultures. However, no unique morphological features of these two isolates were noticed to distinguish them from other Lulworthia spp.
