碩士 / 國立臺灣海洋大學 / 生物科技研究所 / 100 / This research focused on Synechococcus sp. WH7803 which is one of the members of the Cyanophyta genus. The Synechococcus sp. WH7803 was cultivated in two different groups with a light and dark ratio 6:18 and 24:0. Each group was separated into two sub-groups by 14N and 15N environments. The same number from per group of cell was extracted and purified by electrophoresis (SDS-PAGE). Trypsin was added for digesting the protein into peptide for the LC-MS/MS analysis. The data were adopted by SILAC for phycoerythrin identification and quantification of phycoerythrin under various exposure time.
The result showed the phycoerythrin in WH7803 was 4 to 5 times richer in short exposure time. In case of insufficient light, the more expression of phycoerythrin in order to obtain sufficient energy to maintain the physiology of Synechococcus sp.
