Effects of Holothuria atra on exercise performance and anti-aging in senescence accelerated mice / 海參對老化促進小鼠增強運動表現及延緩衰老之研究

碩士 / 靜宜大學 / 食品營養學系 / 100 / Holothuria atra has been shown on reducing lipidemia, anti-hypertension and enhancing immunity, but its effect is indeterminate on exercise performance and anti-aging. The purpose of this study was to investigate the effects of Holothuria atra extracts formula on exercise performance and anti-aging in senescence accelerated mice (SAMP8). Three-month-old male SAMP8 mice were randomly divided into four groups, including control group and Holothuria atra extracts formula group (Low dose 1000mg/kg BW/day, Medium dose 2000mg/kg BW/day and High dose 4000mg/kg BW/day). Each group contained 8 mice. The control group were given double-distilled water (ddH2O) by gavage and the Holothuria atra extracts formula group were given different doses of Holothuria atra extracts formula by gavage for 8 consecutive weeks. The body weight, food intakes and water consumption were measured. The study was divided into two parts: In experiment Ⅰ of increase exercise performance, mices performed on treadmill exercise. The exercise program was performed after trained 0 , 1st , 2nd , 4th and 8th week by all group. During exercise, the electric shock frequencies of exercise program were recorded. The blood glucose, lactate concentration and biochemical parameters in blood were analyzed after the mice were sacrificed. The superoxide dismutase activities (SOD) and malondialdehyde concentration (MDA) of liver in mice were examined. In experiment Ⅱ of anti-aging, the feeding protocol was same in experiment I. In the 7th week, the aging scores were measured, open field activity test, single-trail passive avoidance test and active shuttle avoidance test were recorded in evaluation for learning and memory ability. The mice were sacrificed to analyze the biochemical parameters of sera. The glutathione peroxidase activities (GPx) and MDA of brain in mice were examined. The results showed that there was no significant difference in body weight, food intakes and water consumption among groups. According to results of the exercise program, the retention frequencies of 2nd, 4th and 8th week of Medium and High dose groups were significantly lower than control group (p < 0.05). After running, blood lactate concentration of Medium and High dose groups were significantly lower than control group (p < 0.05). The results showed aging score of Holothuria atra extracts formula group was significantly lower than control group (p < 0.05). Results from single-trail passive avoidance test and active shuttle avoidance test showed that Holothuria atra extracts formula group group was significantly better in learning memory ability (p < 0.05). The anti-oxidation state of liver and brain showed that SOD and GPx activity of Holothuria atra extracts formula group were significantly higher than control group. In addition, the MDA concentration of liver in Holothuria atra extracts formula group were lower than control group (p < 0.05). In summary, the supplement of Holothuria atra extracts formula could increase exercise performance, decrease blood lactate concentration after exercise, promote the antioxidant defense system, and reduce oxidative stress-induced damage. Besides, Holothuria atra extracts formula demonstrated significantly improve learning and memory ability. The underlying increase exercise performance and anti-aging mechanism needs further investigation.
