碩士 / 中國醫藥大學 / 營養學系碩士班 / 101 / This study investigated that the Lentinus edodes root (L. edo root) extract improving Alzheimer''s disease in diabetic animal model. The male wistar rats in this study were divided into five groups:Normal control group (NC); diabetic control group (DM); 0.16、0.48、0.96 g/kgB.W./d of L.edo root in diabetic rats (DM+L、DM+M、DM+H). After six weeks of the treatment, collected blood, liver and brain sample were collected and analyzed. Results showed that DM group significantly increased brain amyloid precursor protein (APP), beta-amyloid 42 (Aβ42), advanced glycation end products (AGEs) and significantly decreased insulin receptor (ISR) levels. L.edo root extract can significantly decreased brain APP, Aβ42, AGEs and significantly increased brain ISR and plasma insulin in diabetes rats. Brain malonaldehyde(MDA), VitE, TNF-α and plasma cholesterol (TC), triglyceride(TG) and alanine transaminase(ALT), MDA were significantly decreased in DM+H group. Elevated plus-maze result show that DM group rats were more anxious. Administration of L. edo root extract for six weeks, improved emotional anxious. In conclusion, biochemical markers of Alzheimer’s disease could be affect by diabetes progression. Insulin resistence maybe highly related to Alzheimer’s disease progression.
