Functional Effect of Phytocompounds from Echinacea purpurea Extract on Zebrafish after Nodavirus infection / 紫錐菊萃取物對斑馬魚抗病毒之功能性研究

碩士 / 弘光科技大學 / 生物科技研究所 / 101 / Echinacea purpurea , commonly known as purple coneflower, is an
important medicinal plant that has been used for centuries, customarily as a
treatment for the common cold, coughs, bronchitis, upper respiratory
infections, and some inflammatory conditions. This study was used
Echinacea purpurea extract (EPE) to enhance immune system to resistance
NNV infected larva zebrafish. Commercial preparations of dried juice were
tested at concentrations ranging from 0.002%,0.02%, 0.04%, 0.08%, 0.2%,
to 2% and compared to aerated water and unstimulated controls. They were
lasting immerse changing every day up to 4 dpf, and observe accumulated
mortality rate, heart rates index and innate immune genes expression(IFN-g,
TNF-a, IL-1b) under EPE treatment. Moreover, we observed zebrafish
accumulated mortality rate, and innate immune genes expression(IFN-g,
TNF-a, IL-1b)by both conditions of pre-treatment EPE then infection NNV
and EPE/-NNV co-treatment. All the data show EPE can modulate these
three genes of IFN-g, TNF-a, IL-1b. The result was indicated the
pre-treatment EPE to decrease fish mortality ratio more obviously about
10~20% than the non-pretreatment EPE fish. Thus, this study demonstrated
that EPE might preclude zebrafish from death owing to trigger innate
immune system to defeat virus infection.
