碩士 / 弘光科技大學 / 化妝品科技研究所 / 101 / Ovatodiolide is one of the index components isolated from the Anisomeles indica. A. indica is a common folk medicinal plant, which has been extensively studied in various diseases, such as gastrointestinal diseases, liver diseases, inflammatory skin diseases and immune system defects. Ovatodiolide is a kind of diterpenoids. It is a bioactive ingredient in A. indica, possesses anti-bacterial and anti-tumor properties, and was traditionally used to treat stomach diseases, inflammatory diseases and hypertension. There are few reports on the application of ovatodiolide as a whitening or antioxidant agents for skin. This study was aimed at investigating the ovatodiolide antioxidant and anti-melanogenic properties of ovatodiolide and assaying the possible mechanisms of action. In the present study, the antioxidant capacity of ovatodiolide were in vitro determined by Folin–Ciocalteau assay, 2,2’-Azino-bis (3-ethylbenzo-thiazline-6-sulfonic acid) (ABTS) radical assay, 2,2-Diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay, reducing power assay and metal chelating ability assay. The results revealed that ovatodiolide has a small reducing power and little total phenolic content. The level of ROS in B16F10 cells was significantly reduced by ovatodiolide. In addition, ovatodiolide inhibited mushroom tyrosinase, reduced the activity of tyrosinase in B16F10 melanoma cells and decreased the melanin content. The Western blot results indicated that ovatodiolide obviously decreased the protein expression levels of tyrosinase, TRP-1, TRP-2 and MITF. Which further block the p-JNK and p-p38 pathway. Our results evidenced that ovatodiolide reduced cellular ROS and inhibited evidenced melanin biosynthesis in B16F10 melanoma cells. Therefore, ovatodiolide may act as the cosmetic whitening and antioxidation ingredient.
